HI my sony gigajuke HI-FI speakers are not working they are plugged into the master output of my VMS2 do I need to tell the virtual DJ software

You can't connect passive (unpowered) speakers directly to the output of a VMS2, or any other device that has a "line level" output.
Passive speakers need to connect to the output of an amplifier. The line level device connects to the input of the amplifier, and the amplifier increases the signal level in order to power the speakers.
